Improved CTAB method for RNA extraction of thick waxy leaf tissues from sago palm (Metroxylon sagu Rottb.)

Abstract Background There is a growing interest in transcriptomics studies parallel to the advancement of transcriptome databases and bioinformatics, which provided opportunity study responses growths, stimuli stresses. an increase demand for excellent RNA extraction techniques. General protocols can be used extraction, but quality quantity vary different types tissues from organisms. Hence, specific method each organism’s tissue type required obtain desired quantity. Results The improved CTAB superior PCI MRIP thick waxy leaves that were applied mature sago palm ( Metroxylon sagu Rottb.) leaf produce total extract with good purity (OD 260/280 ≥ 1.8, OD 260/230 2.0) integrity (RIN ~ 7). sequencing was conducted extracted samples showed assembly results (Q20 97, Q30 91%, mean length 700 bp). Conclusion enables rapid, cost-effective, relatively simple waxy, fibrous high-in-polyphenol M. next-generation recommended quality. Graphical
